Calnexin is a type-I integral membrane protein of the endoplasmic reticulum (ER). It is a calcium-binding chaperone protein that interacts transiently with newly synthesized N-linked glycoproteins, facilitating protein folding and assembly. It may also play a central role in the quality control of protein folding by retaining incorrectly folded protein subunits within the ER for degradation.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Recognizes the D1 domain of an N-glycosylated glycoprotein of 120 kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3 (Leucocyte Workshop VI). CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. Recognizes a protein of 80 kDa-90 kDa, identified as CD36 (Workshop IV; Code P-26). Its epitope maps between aa155-183. It is expressed on platelets, monocytes and macrophages, microvascular endothelial cells, erythrocyte precursors, mammary epithelial cells, and some macrophage derived dendritic cells. CD36 acts as a receptor for thrombospondin (TSP), collagen types I, IV and V, P. falciparum malaria-infected erythrocytes, and sickle erythrocytes. It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN). CD36 plays a role in platelet aggregation, macrophage foam cell development, inflammation, and the tissue ischemia observed in sickle cell disease and cerebral malaria. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Epitope of GZ-1 and GZ-20 monoclonal antibodies are different. EGP40 is a 40-43 kDa transmembrane epithelial glycoprotein, also identified as epithelial specific antigen (ESA), or epithelial cellular adhesion molecule (Ep-CAM). It is expressed on basolateral cell surface in most simple epithelia and a vast majority of carcinomas. Recognizes a protein of 185 kDa, which is identified as c-erbB-2/HER-2/neu. Its epitope is localized in the extracellular domain. C-erbB-2/HER-2 is a member of the EGFR family. This MAb is specific and shows minimal cross-reaction with other members of the EGFR-family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. Biotium Cytokeratin 19(Ks19.1), CF488A conjugate, 0.1mg/mL
This MAb reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40 kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Biotium Cytokeratin, LMW(AE-1), CF488A conjugate, 0.1mg/mL
This MAb recognizes the 56.5 kDa (CK10); 50 kDa (CK14); 50 kDa (CK15); 48 kDa (CK16); 40 kDa (CK19) keratins of the acidic (Type I or LMW) subfamily. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50', 48, 46, 45, and 40 kDa. MAb AE3 recognizes the 65-67, 64, 59, 58, 56, and 52 kDa keratins of basic subfamily. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. Biotium CD81 / TAPA-1 (1.3.3.22), CF488A conjugate, 0.1mg/mL
This antibody recognizes a protein of 26 kDa, identified as CD81 (Workshop VI; Code CD81.1). CD81 has a very broad cellular distribution, being expressed on T- and B-lymphocytes, NK cells, thymocytes, eosinophils, fibroblasts, epithelial and endothelial cells. Neutrophils, erythrocytes and platelets are negative, while monocytes are variably positive. CD81 is a member of a family of tetraspanin transmembrane proteins, including CD9, CD37, CD53, CD63, and CD82. It associates with CD19, CD21, Leu 13, and integrins on cell membrane and is involved in signal transduction in B lymphocyte development and cell adhesion. CD81 also acts as a receptor for the envelope protein E2 of chronic hepatitis C virus. CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 beta chain (CD8 alpha/beta) or as a homodimer (CD8 alpha/alpha). A majority of thymocytes and a subpopulation of mature T cells and NK cell express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed.
